What Is Speciogynine?

Speciogynine is a diastereomer of mitragynine and typically the third- or fourth-most abundant alkaloid in kratom, comprising roughly 7–21% of total alkaloid content. Like paynantheine, speciogynine acts primarily as a smooth muscle relaxant rather than an opioid receptor agonist. It has negligible activity at the mu-opioid receptor and does not contribute meaningfully to kratom's analgesic or euphoric effects. Its smooth muscle relaxant properties may contribute to some of the gastrointestinal and vascular effects reported by kratom users. As a diastereomer of mitragynine (same molecular formula, different spatial arrangement), speciogynine demonstrates how small structural differences can result in dramatically different pharmacological profiles. Research into speciogynine remains limited, and its precise role in kratom's overall effect profile is still being characterized.

Dose-Dependent Effects

Receptor Activity

ReceptorKi (nM)Activity TypeWhat This Means
Mu-opioid (MOR)NegligibleDoes not meaningfully activate opioid receptors despite being a structural relative (diastereomer) of mitragynine.
Smooth muscleRelaxantRelaxes vascular and gastrointestinal smooth muscle. This is considered its primary pharmacological action, similar to paynantheine.

Safety & Adverse Effects

Research status

Speciogynine is an early-stage research compound with limited human pharmacokinetic data. It is present in moderate-to-high concentrations in kratom but does not appear to drive the plant's opioid-like effects. Its smooth muscle relaxant activity may contribute to vascular and GI effects.

Drug Interactions

Antihypertensives and vasodilators

moderate

Theoretical additive blood pressure lowering due to smooth muscle relaxant properties. Clinical significance at typical kratom doses is not established.

CNS depressants (benzodiazepines, alcohol, opioids)

major

As a component of whole-leaf kratom, speciogynine is consumed alongside opioid-active alkaloids. Standard kratom interaction warnings apply.
